Melanotan 2 (MT-2) is a lab-made version of alpha-melanocyte-stimulating hormone, which was first synthesized in the 1980s. MT-2 has been found to have several effects, such as increasing sexual arousal, reducing compulsive and addictive behaviors, suppressing appetite, and promoting lean body mass. Studies have shown that MT-2 stimulates melanocytes, leading to increased skin pigmentation, and it may also have potential benefits in early childhood development for autism.
Initially created as a sunless tanning option, MT-2 was discovered to have a range of effects due to its ability to bind with melanocortin receptors. There are five known melanocortin receptors, and MT-2 primarily binds with MC-4R and MC-1R, which are involved in feeding, sexual behavior, and skin pigmentation.
Research has indicated that MT-2 may help reduce fat storage and hunger in animals by acting as a potent agonist of the MC-4R receptor, which plays a role in food intake and preference. MT-2 has also been shown to stimulate both leptin-dependent and leptin-independent pathways, making it a more effective treatment for reducing hunger than leptin alone. Furthermore, MT-2 may increase thyrotropin-releasing hormone expression, which is associated with satiety and food intake.
Although MT-2 has been investigated in clinical trials, challenges with its administration have led to ongoing research into its benefits. The peptide exhibits minimal to moderate side effects, has low oral bioavailability, and excellent subcutaneous bioavailability in mice.
